Description

Principle of the assay: human Ficolin-3 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for Ficolin-3 has been precoated onto 96-well plates. Standards(NSO, Q24-R299) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Ficolin-3 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human Ficolin-3 amount of sample captured in plate.
Background: Ficolin-3, also known as HAKA1, is a protein that in humans is encoded by the FCN3 gene. It is mapped to 1p36.11. The protein can activate the complement pathway in association with MASPs and sMAP, thereby aiding in host defense through the activation of the lectin pathway. Ficolin-3 is expressed as a 35-kD protein that reacts with systemic lupus erythematosus sera. Ficolin-3 does not bind fibronectin (FN1), elastin (ELN), or zymosan, and the lectin activity of the Ficolin-3 is calcium independent.
